Abstract:

The involvement of M2- like tumor assosiated macrophages TAM? in the advancement and treatment of cancers has been widely documented. The objectives of this study was to develop a new signature associated with M2- like TAMs to fore cast the prognosis and tratment response individuals diagnosed with breast cancer (BC)
Methods: Weighted gene co-expression network analysis(WGCNA) was used to screen for M2-like TAM-related modular genes. The TAM-related modular subtype was then identified through unsupervised clustering. A predictive model was created using the genes that were expressed differently in the two clusters. To further uncover the immune environment of BC patients, investigations were conducted on immune cells, TME score, and response to immunotherapy in two different risk groups.
Results: WGCNA identified a total of 722 M2-Like TAM genes, out of which 204 genes were found to be related to RFS. Two clusters were identified and patients with cluster I exhibited upregulated cancer-related pathway, a higher proportion of triple -negative breast cancer (TNBC) subtype, lower expression of immune checkpoints, and worse prognosis. Cluster 2 was characterized by the upregulated immune-related pathways, higher proportion of luminal A subtype, and higher expression of immune checkpoints. Subsequently, a prognostic signature was created and confirmed in an independent dataset. By utilizing a risk score and clinical characteristics, a well-built nomogram can accurately forecast the survival outcome for every individual with utmost stability. Furthermore, patients classified as low-risk exhibited a more favorable outlook, e levated TME score, and a superior reaction to immunothepapy.
Conclusions: We discovered two different types of M2-like TAMs and developed a prognostic signature, revealing the diversity of M2-like TAMs in BC and their correlation with immune status and prognosis. This feature can predict the prognosis and immunotherapy effect BC, and offer novel concepts and approaches for tailoring the treatment of BC.

Biography:

Yuan Zhang studied oncology at the Forth Military Medical University, xian, shannxi,china and graduated as MS in 2013, I reciebed my PHD degree in 2016. I then worked in Shaanxi Provincial People’s Hospital, Xian, 710068, Shannxi, as an oncologist from 2018 to now. I had published about seven research articles in SCI journals, my reseach focus on BC resistance and the Drug resistance mechanism.
