Title : Multiple applications of withaferin A and study of its biosynthesis in the withania somnifera: A comprehensive approach

Abstract:

Withania somnifera (WS), a significant medicinal plant utilized in the Ayurvedic system, is a rich source of withanolides, which are pharmacologically active secondary metabolites. Withanolides, withaferin-A, withanolide-A, withanoside-IV, and certain sitoindosides are the primary constituents of WS. Withaferin A, which has anti-inflammatory, anti-invasive, and anti-angiogenic properties, is the most bioactive of these substances. In light of contemporary pharmacology, it is important to comprehend the fundamental mechanism underlying the wide spectrum of bioactivities induced by Withaferin A. Therefore, much work was put into determining Withaferin A's function intracellularly and characterizing its target proteins. Mevalonate (MVA) and methyl-D-erythritol-4-phosphate (MEP) pathways, both of which utilize 24-methylene (C30-terpenoid) cholesterol as a metabolic intermediary, are used to synthesize withanolides. Enzymes needed for the manufacture of terpenoid backbone are encoded by several putative genes. In this review, we will explore the molecular foundation for the possible health benefits of withaferin A as well as the biosynthesis of withanolide. This study will also provide insight on the neuroprotective properties of withaferin A and withanolide synthesis and help create new techniques for the functional genomics of this significant medicinal plant.
